Melanotan II: A Research Reference for Melanocortin Signalling

Melanotan II is a synthetic cyclic heptapeptide analogue of α-melanocyte-stimulating hormone (α-MSH). It engages multiple melanocortin receptors (MC1R, MC3R, MC4R, MC5R) and has been used as a research reference for melanocortin-pathway pharmacology since its development in the 1980s.

MC1R is the most studied receptor in the context of pigmentation. Activation drives the eumelanin synthesis pathway in melanocytes, and Melanotan II provides a useful in-vitro tool for studying these signalling cascades. MC4R has attracted separate research interest for its role in appetite regulation and sexual-function signalling.

In pharmacology research, Melanotan II's non-selective engagement of multiple MC receptors makes it useful for mapping receptor-specific effects when paired with selective antagonists. More selective analogues such as Bremelanotide (PT-141) are then used to isolate MC4R-specific responses.

From a practical research perspective, Melanotan II is supplied lyophilised. Reconstitution in bacteriostatic water gives a stable solution. The peptide is generally stable when stored cold and protected from light. As always, reference vials should ship with HPLC purity verification and a batch certificate of analysis.

Comparative research designs frequently pair Melanotan II with α-MSH itself, with NDP-MSH (a more potent analogue) and with receptor-selective compounds. This kind of structure-activity work continues to inform medicinal-chemistry programmes around the melanocortin family.
