Peptide Reconstitution and Storage: A Laboratory Reference

Research-grade peptides ship lyophilised because the dry, low-moisture state preserves chemical integrity far better than any liquid formulation. Correct reconstitution is the first laboratory step that determines whether the assay reflects the molecule's true pharmacology or an artefact of degradation.

Bacteriostatic water (sterile water with 0.9% benzyl alcohol) is the most common reconstitution solvent for peptides intended for repeated sampling, because the preservative discourages microbial growth. Sterile saline or sterile water is appropriate for single-use preparations. Highly hydrophobic peptides may require small amounts of acetic acid or DMSO for full dissolution — solvent compatibility with the downstream assay must be checked first.

Aliquoting is essential. Freeze-thaw cycles are one of the largest sources of peptide degradation in chronic laboratory studies. Reconstituting a vial, then immediately splitting it into single-use aliquots, eliminates repeated freezing and thawing of the bulk sample.

Storage temperature depends on the time horizon. Lyophilised peptide stored at -20 °C is generally stable for years. Reconstituted peptide stored at 4 °C is typically used within a documented stability window — frequently two to four weeks for stable peptides, less for fragile sequences. Reconstituted aliquots stored at -80 °C extend usable life considerably.

Documentation is the often-overlooked step. Every reconstitution should be recorded: date, solvent, concentration, volume, peptide lot, freezer location. This trail supports reproducibility and is essential when publishing or replicating prior work.
